Usage of ni and no (Spanish)

I heard this:

Not at a quarter to three = Ni a las tres menos cuarto
And later the speaker said Ni a las tres y cuarto.
So he mentioned 2 times on which he couldn't come.

My question: when using ni, should there always be at least 2 things you mention? 2 negative things?
Or could you also simply use it as a "no".
When someone asks you for instance if you can come at three you reply: Ni a las tres.
Or would you then say "A las tres no"?

So when do you use "ni" and when ".... no"?
If I understand correct ni means neither, so that must involve more no's. So in this last example I should use no, correct?

Correct.

If someone asks you if you can come at three you could reply:
"Si puedo" = I can
OR
"No puedo" = I can't
OR
"Ni a las tres ni a las cuatro" = Nor at three nor at four
